What is 5-Amino-1MQ?

5-Amino-1MQ is a small-molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT), not a peptide, sold online as a research chemical for weight loss. NNMT regulates how cells balance energy and methylation, and its abnormal activation is linked to diabetes, obesity, fibrosis, and cancer. 5-Amino-1MQ is not FDA-approved and does not appear on FDA's list of bulk substances nominated for compounding.

What does 5-Amino-1MQ do?

5-Amino-1MQ is not a peptide; it is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme a 2026 review describes as helping "regulate how cells balance energy and methylation potential" [1]. The same review says abnormal NNMT activation can disrupt that balance, "fueling diseases such as diabetes, obesity, fibrosis, and cancer" [1]. It notes that small-molecule NNMT inhibitors as a class have had "limited preclinical success" from "insufficient target engagement, reduced bioavailability, and unknown safety profiles," though newer compounds show "promising safety profiles" in preclinical work [1].

What does the research show, and is it FDA-regulated?

The evidence is from animals only. In an 11-day study, obese mice on a high-fat diet that were given 5-Amino-1MQ lost body weight and white fat mass and had lower plasma total cholesterol, with no change in food intake [4]. A different NNMT inhibitor produced similar results in mice with diet-induced obesity: it "caused a reduction in body weight, improved insulin sensitivity and normalized glucose tolerance," an effect absent in mice lacking the NNMT gene [2]. 5-Amino-1MQ does not appear in FDA's May 2026 list of bulk substances nominated for compounding under section 503A, and it is not an FDA-approved drug [3].
