What is GLP-2?

GLP-2 is a gut hormone released from intestinal L cells that promotes intestinal growth, absorption and blood flow, distinct from GLP-1, the hormone targeted by weight-loss drugs. Researchers derived Gattex (teduglutide), an FDA-approved GLP-2 analog, from this biology to treat short bowel syndrome.

What does GLP-2 do in the body?

Glucagon-like peptide-2 is an intestinally derived hormone released from intestinal L cells that enhances intestinal growth, digestion, absorption, barrier function and blood flow [1]. It also aids repair after intestinal damage or bowel resection, and its effects on the gut are largely mirrored in humans with intestinal failure [1]. GLP-2 acts through its own GLP-2 receptor, which is concentrated almost entirely in the intestinal tract [1].

How is GLP-2 different from GLP-1?

GLP-2 is liberated from the proglucagon precursor, secreted by intestinal L cells, and acts through a receptor that is highly localized to the intestinal tract [1]. GLP-1, covered on its own page, works through a different receptor and is the basis for medications like Ozempic and Wegovy. GLP-2's gut specificity led instead to a therapy for short bowel syndrome, not for diabetes or obesity [1][2].

Is there an FDA-approved GLP-2 drug?

Yes. Gattex (teduglutide) is an FDA-approved GLP-2 analog indicated for adults and children age 1 and older with short bowel syndrome who depend on parenteral support [2]. It is given as a once-daily subcutaneous injection and works by activating intestinal GLP-2 receptors to increase blood flow and nutrient absorption [2]. Gattex is unrelated to GLP-1 weight-loss drugs.
